What is PI3K?

The PI3K (phosphatidylinositol 3-Kinase) pathway is one of the most commonly activated cell signaling pathways in human cancers. When it functions normally, the PI3K pathway regulates key cell functions such as growth, motility, proliferation, differentiation, survival and intracellular messaging. But, when problems occur in the pathway, the resulting dysregulation of these cell functions can contribute to the development of many different types of cancers, including breast, ovarian and endometrial cancers.

Problems with the PI3K signaling pathway have an influence on cell growth and division. When cell growth and division are out of control, it can result in the formation of tumors. Such problems are caused by mutations in PI3K and other genes in the pathway. Once mutations occur, new cells which result from cell growth and division also carry the mutations. The unregulated growth in effect “snowballs” to the point where cancer can develop.

Current research is focused on how to control cancer cell growth by dampening abnormal PI3K signaling. Although the mutations cannot be corrected, their influence can be controlled and regulated by compounds (drugs) known as PI3K inhibitors. By targeting the signaling pathway, these inhibitors can block signals from PI3K and slow down or eliminate the out of control growth of mutated cells.
